Melatonin significantly reduced sleep onset latency by 7.06 minutes (p<0.001), increased total sleep time by 8.25 minutes (p=0.013), and improved overall sleep quality (p<0.001) versus placebo. Higher doses and longer durations showed greater effects.

Dose-response meta-analysis of 26 RCTs (n=1,689) optimizing melatonin as a sleep-promoting agent. Sleep onset latency reduction and total sleep time increase peaked at 4 mg/day. Systematic review and meta-analysis of 60 studies (n=6,894) on valerian for sleep and associated disorders. Meta-analysis of 10 placebo-controlled RCTs found a combined effect size of 0.36 for subjective sleep quality improvement. Subgroup analysis favored whole root preparations (ES=0.83) over extracts (ES=0.10). No severe adverse events reported.
