The most-cited study enrolled 30 Japanese adults aged 50โ80 with mild cognitive impairment and randomized them to 3g/day of lion's mane fruiting body extract or placebo for 16 weeks. The lion's mane group showed significantly higher scores on cognitive assessment from week 8 onward. Effects reversed within four weeks of stopping โ suggesting the benefit requires continued use.2 A meaningful positive finding in a population with mild cognitive impairment, though extrapolating these results to healthy adults in their 30s requires caution.
A 2019 study in healthy older adults found improvements in cognitive function, sleep quality, and mood scores after 12 weeks at 3g/day โ a broader outcomes picture suggesting the compound may act through more than one pathway.3
A third RCT in younger healthy adults (n=41, 12 weeks, 1.8g/day) found improvements specifically in the Stroop Color-Word Test โ a validated measure of cognitive flexibility and executive function โ but not across all cognitive domains tested.4 Mixed results in a small trial are informative rather than definitive, but they do establish that some cognitive benefit is observable in healthy, non-impaired adults at a research-accessible dose.
HealthDataConsortium's 2026 review summarized the completed human trials as each reporting "at least one positive cognitive outcome." That framing is accurate and appropriately humble โ all three trials showed something positive, but the evidence base remains limited by sample size and follow-up duration. The Alzheimer's Drug Discovery Foundation's current rating is "promising/insufficient evidence," which captures the 2026 state of the science well.
